TotalflavonoidofLitseacoreanaleveexertsanti-oxidativeeffectsandalleviatesfocalcerebralischemia/reperfusioninjuryShuyingDong1,2,XuhuiTong2,JunLi1,ChengHuang1,ChengmuHu1,HaoJiao2,YuchenGu21SchoolofPharmacy,AnhuiKeyLaboratoryofBioactivityofNaturalProducts,AnhuiMedicalUniversity,Hefei230022,AnhuiProvince,China2FacultyofPharmacy,BengbuMedicalCollege,Bengbu233030,AnhuiProvince,ChinaAbstract:Inthisstudy,wehypothesizedthattotalflavonoidofLitseacoreanaleve(TFLC)protectsagainstfocalcerebralischemia/reperfusioninjury.TFLC(25,50,100mg/kg)wasadministeredorallytoaratmodeloffocalischemia/reperfusioninjury,whilethefreeradicalscavengingagent,edaravone,wasusedasapositivecontroldrug.Resultsofneurologicaldeficitscoring,2,3,5-triphenyltetrazoliumchloridestaining,hematoxylin-eosinstainingandbiochemicaltestsshowedthatTFLCatdifferentdosessignificantlyalleviatedcerebralischemia-inducedneurologicaldeficitsandhistopathologicalchanges,andreducedinfarctvolume.Moreover,itsuppressedtheincreaseinthelevelsofnitratesplusnitrites,malondialdehydeandlactatedehydrogenase,anditdiminishedthereductioningluta-thione,superoxidedismutaseandcatalaseactivitiesinducedbycerebralischemia/reperfusionin-jury.Comparedwithedaravone,theprotectiveeffectsofTFLCatlowandmediumdoses(25,50mg/kg)againstcerebralischemia/reperfusioninjurywereweaker,whiletheprotectiveeffectsathighdose(100mg/kg)weresimilar.OurexperimentalfindingssuggestthatTFLCexertsneuroprotectiveeffectsagainstfocalcerebralischemia/reperfusioninjuryinrats,andthattheeffectsmaybeasso-ciatedwithitsantioxidantactivities.Keywords:neuralregeneration;totalflavonoidsofLitseacoreanaleve;focalcerebralischemia/reperfusioninjury;oxidativestress;neuroprotection;infarctvolume;neurologicaldeficitscores;malondialdehyde;glutathione;superoxidedismutase;grants-supportedpaper;neuroregeneration;Acceptancedate:2013-09-05Citethisarticle:ShuyingDong,XuhuiTong,JunLi,ChengHuang,ChengmuHu,HaoJiao,YuchenGu.TotalflavonoidofLitseacoreanaleveexertsanti-oxidativeeffectsandalleviatesfocalcerebralischemia/reperfusioninjury[J].NeuralRegenerationResearch,2013,8(34):3193-3202.
