广东药学院硕士学位论文枇杷叶质量评价的研究姓名:韩秀奇申请学位级别:硕士专业:中药学指导教师:李书渊2011-05I11.2850IIiStudiesonthequalityassessmentofEriobotryaeFoliumHanXiuqiTraditionalChineseMedicineSupervisor:ProfessorLiShuyuanEriobotryaeFoliumisthedriedleafofEriobotryajaponica(Thunb.)Lindl.(Fam.Rosaceae),whichisatraditionalChinesemedicinerecordedineachversionsofChinesePharmacopoeia.Ittastesbitterandpungent,slightlycoldinnatureandbelongstotheLungMeridianandstomachmeridian,toremoveheatfromthelungandthestomach,andtorelievecoughandvomiting.Pharmacologicalstudieshaveshowngoodanti-inflammatoryandcougheffectoftheEriobotryaeFolium.SomerecentresearchesalsofoundthatEriobotryaeFoliumhaveothereffectssuchasanti-tumor,antiviral,hypoglycemic,hepatoprotectiveandcholeretic,scavengingoxygenfreeradicals,enhanceimmunefunctionandsoon.EriobotryaeFoliumisrichinresourcesinChina,mainlyintheSouthRegionandShaanxi,Gansu,Jiangsu,Anhui,Zhejiang,Jiangxi,Guangdong,Fujian,Taiwan,Sichuan,Guizhou,Yunnanandotherplaces.Cultivationareais112,800hm2,accountingformorethan50%ofworldproduction.EriobotryaeFoliumcouldbeharvestedthroughoutayear,butnostrictstandardsontheharvestingtime.ThemainactiveingredientsofEriobotryaeFoliumareflavonoidsandtriterpeneacids.TheflavonoidsinEriobotryaeFoliumareaseriesofcompoundswithmother-nucleusofquercetinandkaempferol,butcontentislow.Inthisstudy,flavonoidsinEriobotryaeFoliumwashydrolysised,quercetinandkaempferolwereasthetargetcompoundsofthecontentofflavonoids,andhaveestablishedthedeterminationmethodofflavonoidsofEriobotryaeFolium.Thisstudyhavecomparedthecontentsoftotalflavonoids,quercetinandkaempferolofEriobotryaeFoliumcollectingfromdifferentregionsanddifferentmonths.Theexperimentsdeterminatedthecontentofquercetinandkaempferolafterhydrolysis,thencalculatingthetotalflavonoids,thetotalflavonoidscontentsdiffersignificantlyindifferentmonths,thecontentsofsamplesinAprilandMaywerehigh,andinthemonthofJuly,August,Septemberalsohigh,andMarchwastheiilowest.ThefloweringseasonisthemonthsofSeptembertoNovemberandthefruitingseasonisthemonthsofApriltoMayinthefollowingyear,andcontentoftotalflavonoidsinthemonthsofSeptember,OctoberandthemonthsApril,Mayarehigher,andrelativelylowcontentinthemonthsofMarch,June,November.Itprovesthatcontentsofflavonoidswithfloweringandfruitingmayexistcertaincorrelation,buthowtherelationshipbetweenthemshouldlookforwardtothefurtherstudy.EriobotryaeFoliumfromdifferentregions,thequercetinandkaempferollevelsaredifferentmaybebecauseoftheenvironment,growthtime,pickingtimeandotherfactors.ThisstudyhavemeasuredcontentsofthefourbatchesofEriobotryaeFoliumsamplesbeforeandafterprocessing(Paozhi).Afterprocessing,thequercetin,kaempferolandtotalflavonoidsarelessthanbefore,andofwhichquercetinisdecreasedinthelargestproportion,probablypost-processingthechemicalstructureofflavonoidswaschangedsothecontentresultedinlower.ThisstudywasfirstbuildthemethodinthesimultaneousdeterminationofcontentofquercetinandkaempferolinEriobotryaeFoliumwhichisaccurateandreliable,reproducible,strongfeatures,simpleandsoon.Inrecentyears,alargenumberofstudieshavefoundthattheursolicacidandoleanolicacidinEriobotryaeFoliumhassignificantanti-tumorandboosttheimmunefunctionbesideanti-inflammatory,safeguardliverandanalgesic.InthequalitycontrolstandardsofEriobotryaeFolium,Thecontentsofoleanolicacidandursolicacidweredeterminatedin“ChinesePharmacopoeia”(2010edition,part).Thisexperimenthasshowthattheoleanolicacidandursolicacidcontentarechangebythedifferentseasons,whilethecontentofursolicacidhaschangelittle.Andthecontentsofoleanolicacidandursolicacidarealsoalittledifferentfromthedifferentregions.Thecontentsofursolicacidandoleanolicacidhavedecreasedafterprocessing,butnotverysignificant.Inthisstudy,PharmacologyexperimentsforrelievingcoughandasthmaandreducingsputumhavebedonebythesamplesincludethewaterextractandalcoholextractbothrawandprocessingEriobotryaeFolium,andtheflavonoidfractionandtriterpenefraction.Theresultsshowthatthewaterextractisbetterthanthealcoholextract.AndtheeffectoftheprocessingFoliumEriobotryae’swaterextractisthebest.Comparedwiththecontrolgroup,theflavonoidfractionandtriterpenoidfractionalsohavetheeffectsofcoughandasthma.Thecontentsofursolicacidandoleanolicacidwereslightlydecreasedafterprocessing,whiletheflavonoidwasdecreasedbynearlyahalf.ButthepharmacologicaliiiexperimentsshowthatffectiveofprocessingEriobotryaeFoliumismuchbetterthanrawEriobotryaeFoliumonthecough,phlegmandasthma.Thereareothercompoundsthatworkinpharmacologicalaction.Keywords:EriobotryaeFolium;qualityevaluation;totalflavonoids;HPLC;coughandasthma1hyptadienicacid[4]2a3b,6a,19a[6],tormeticacid,1b-hydroxyeuscaphicacid[7],3b,6b,19a-trihydroxyurs-12-en-29-oicacid,23-Trans-p-coumaryltormenticacid,23-cis-p-coumaryltormenticacid,3-o-trans-caffeoyltormenticacid,3-o-trans-p-coumarylrotundicacid[8],3-o-trans-feruloyleuscaphicacid[9]-3-O-[10],quercetin-7a-L-rhamnosidequercetin-3-o-sophorosidequercetin-3-o-rutinoside,quercetin-3-rhamnoside,kaempferol-3-o-glucoside,kaempferol-3-ogalactoside,kaempferol-3-o-nehes-peridoside,Kaempferol-3-o-rhamnoside,kae
