EffectResearchofImmobilizedAlgae-bacteriaRemov-alAmmoniaNitrogenofAquacultureWastewaterandProposedModelZOUWan-sheng1,2,ZHANGJing-lai2,LIULiang-guo1,DENGWu-jun31.LifeSciencesofArtsandsciencesofHunanUniversity,Changde415000;2.SchoolofenvironmentalSciencesRenminUniversityofChi-na,Beijing100872;3.InstituteofSubtropicalAgriculture,ChineseAcademyofSciences,Changsha410000Abstract AppliedImmobilizedalgaebacteria(ABI)toremoveammoniaoffreshwateraquaculturewastewater.Temperature(T),PH,lightin-tensity(I),dissolvedoxygen(DO)andfillingratefivefactorsplaysimportantroleintheprocessofammonianitrogenremoval,relateddatabe-tweenammoniaremovalandfivefactorswasreceivedthroughmulti-factororthogonaltest,andestablishedrelationsmodelbetweenthefivefac-torandnitrogenremoval.Theresultsshowthatfive-factorshadsignificanteffectonAR,andthebestcombinationsforremovingARwastemper-ature30℃,pH=7.0,lightintensity6000lux,dissolvedoxygen5.0mg/Landthefillrate10%.Accordingtotheexperimentaldata,equationmodelwasproposedandcoefficientofdeterminationR2=0.8648,P0.05.SamplesT-testwasdonebetweenthemodelpredictionsandtheactualmeasuredvalues.Testresultsshowedthatthesignificantdifferenceofoverallmeanvaluesig.(2-tailed)was0.978(P0.05),itShowsthathadnosignificantdifferencebetweenmodelpredictionsandtheactualmeasuredvalue,andmodelhadahighdegreeoffitting.Keywords ImmobilizedAlgae-bacteria;Aquaculturewastewater;Ammoniaremoverate;ProposedmodelReceived:May24,2010  Accepted:July1,2010SupportedbytheNationalNaturalScienceFoundationofChina(No.30972260).*Correspondingauthor.E-mail:zwsksy@163.com  Algae-bacteriaImmobilized(ABI)isthemixtureofspecif-icalgaeandmicrobialbacteria.Itisaphycomycetescollabo-rativesymbioticimmobilizedsystemwhichisformedbygel,crosslinkingagentandphycomycetes[1-4].Becauseofhighbiomassconcentration,easysolid-liquidseparation,strongtolerance,fewexcesssludges,itiswidelyusedforwastewatertreatment,purificationandseparatingpollutantsfromwastewater[5-7].However,whenimmobilizedalgal-bac-terialisusedtotreatwastewater,thenremovaleffectsarein-fluencedbymanyexternalfactors,particularly,duringlarge-scaletreatment,theinfluentialfactorsarenotallcontrolled.Olguinetal[8]havebelievedthatundercertainremovalrateofpollutans,therearesubstitutioneffectsamongdifferentinflu-entialfactors,namley,therearedifferentconditionalcombina-tionsofdifferentinfluentialfactorswhichcangeneratethesameremovaleffectonpollutants.Therefore,theABItreatedammonianitrogenofandAmmonia-removalRate(AR)aretakenasmainindexes.Throughsettingtheinfluencesofin-vestigationfactorsoforthogonalexperimentonAR,suchastemperature,pH,lightintensity,dissolvedoxygen,fillingrate,thecombinedmodelisconstructed.MaterialsandMethodsMaterialsExperimentalmaterialsandculture Chlorellapyrenoinoid-osawereexperimentalmaterialsandthealgaespecieswereprovidedbyInstituteofHydrobiology,ChineseAcademyofSciences.TheculturemediawereKuhlculturemedia[9]andmicrobialbacteriawerereplacedbyactivatedsludge,whichwasprovidedbySewageTreatmentCenterofChangdeCity,HunanProvince.Underasepticmanipulationcondition,Chlo-rellapyrenoinoidosawereinnoculatedinculturemedia,thentheywereconductedamplificationcultureinbiochemicalincu-batorattheconditionof25℃,5000lxlightintensityand12∶12light-darkratio.Selectionandpreparationoffreshwateraquaculturewastewater Thefreshwateraquaculturewastewaterwascol-lectedfromthepondBaimaLakeBreedingFishBase,ChangdeCity.Thewastewaterwasfirstlyfilteredthrough0.45μmcellu-loseacetatemembraneandthewastewaterqualityswere310.8mg/LCODcr,17.3mg/LNTU,31.0mg/LammonianitrogenandpH7.2.CODdetectionwasaccordingtopotassiumdichromatemethod,NTUwasdectectedbycommonspectrophotometry,ammonianitrogenwasdetectedbyNesslersreagentsspectro-photometerandpHwasdetectedbypHcompositeelectrode.OtherchemicalreagentswereaccordingtoKuhlculturemediaformulaandthenitrogensourcewasonlyreplacedbyammonianitrogeninwastewater.MethodsFixationmethodsofphycomycetes ThefixationmethodwasMix-immobilization(MI)[10-14]:theculturedChlorellapyre-noinoidosawasconducted2500r/mincentrifugalconcentra-tionandthesupernatantswereabandoned.Thealgalcellsconcentratedsolutionandconcentratedsolutionoffreshacti-vatedsludgeweremixedwiththesamevolumn,thentheyweremixedwithsterilizedgel(2%sodiumalginateSA+8%polyvinylalcoholPVA)toformamixturewith2%volumeratio.Acertainamountofmixturewasdropptedintoprecoo-lingcrosslinkingagent(2%CaCl2+4.5%H3BO3)solutiontoformphycomycetesimmobilizedrubberball.Therubberballwasplaced4hinsolutionforfutureuse.Experimentaldesignandmethod Temperature(A),pH(B),lightintensity(C),dissolvedoxygen(D)andfillingrate(E)werechosentoconductorthogonaltest.Beforeexperi-ment,allfactorswereoptimizedtomakeallfactorsintherangeofphycomycetesnormalgrowth.Thetestingfactorlev-AgriculturalScienceTechnology,2010,11(5):117-120Copyright2010,InformationInstituteofHAAS.Allrightsreserved.ResourcesandEnvironmentelwasdesignedaccordingtoL25(5∧5).Table1 TheorthogonaltestoffivefactorsLevelsFactorsTempera-ture∥℃(A)pHvalue(B)Lightinten-sity∥lx(C)Dissolvedoxygen∥mg/L(D)Fillingrate∥%(E)One206.020002.05Two
