广西师范大学硕士学位论文荸荠组织培养研究姓名:潘登浪申请学位级别:硕士专业:野生动植物保护与利用指导教师:李伯林20070501I2004(1)6-BAKTNAAPH6-BAKT6-BANAAKTKT00.5mg/L)NAA(0.010.2mg/L)6-BA0.5mg/L+KT0.5mg/L+NAA0.01mg/L4.322cm2.12PHPH4.017.00PHpHpH6.00(4.32)2.121832254.322.1225MS+6-BA0.5mg/L+KT0.5mg/L+NAA0.01mg/L+3%+0.7%25pH6224-D6-BAKTZT24-D6-BAKTZT:MS+2,4-D1.0mg/L+6-BA0.5mg/L+3%+0.65%88.19%II0Lux120Lux2000Lux24-DTDZTDZ3MS+IBA0.3mg/L+0.01%+3%100%154+9,+,100%,530.4%64.2%10%9.91%6-BA6-KTZTTDZLFSNAAIBA24-D24-IIITheTissuesCultureStudyofEleocharistuberosa(Roxb)Roemet.SchultThesisauthor:PanDenglangSupervisor:LiBolinMajor:WildlifeprotectionandusenessGrate:2004AbstractThepropagation,growthandrootsinductionofChinesewaterchestnut[Eleocharistuberosa(Roxb)Roemet.Schult]clusterbudswerestudiedinthispaper.Thecallusinductionanddifferentiation,theeffectsofdifferentfactstothesurvivalandgrowthofcultureseedlingswerealsoexperimented.Theyieldandqualityofcormwerecomparedbetweencultureseedlingsandtraditionalseedlings.ThisresearchcanenrichthecontentoftissueculturerapidpropagationsystemandbuildupthecormquailtyestimatesystemofChinesewaterchestnut(estimateindex:products&sizeclassification,appearancecharacter,innerphysical&chemicalcharacter),andtoprovidethegistforchoicenessspecieschosen,seedlingsproduceandproductionexploitation.Theresultsareasfollows:(1)Fortheclusterbuds,theculturemediumandconditionhasamoretremendousinfluencetothepropagationcoefficientandthegrowthfettle.Theinfluencesoftheplantgrowthregulatorscompounding,pHandtemperaturewerestudied.Theresultsshowedthat:6-BAhadaremarkablefunctiontopromotethepropagationofbuds.Butastofoliarstem,itshowedanotablerestrainfunctionalongwithitshigherconcentration.Ithadthebiggestfunctionamongthethreekindsofplantgrowthregulators.Astopropagationcoefficient,thefunctionofKTwasinferiorto6-BA,andstrongerthanNAA.Forthegrowthoffoliarstem,KThadtheweakestfunctionofthethreehormones.ThefunctionofKT(concentrationis0~0.5mg/L)andNAA(concentrationis0~0.5mg/L),tothepropagationcoefficientofclusterbudsandthegrowthoffoliarstem,showedarulethatitwouldweakenfirstandthenincreaselateralongwiththeconcentrationincreased.Colligatingpropagationcoefficientofclusterbudandgrowthfettleoffoliarstem,thefavorablecompoundingofplantgrowthregulatorswas:6-BA0.5mg/L+KT0.5mg/L+NAA0.01mg/L.Itspropagationcoefficientofclusterbudsreached4.32andtheaveragenumberoffoliarstemsperfleshystem(lengthlongerthanorequal2cm)was2.12.ThepropagationcoefficientandthegrowthfettleweredifferentwhentheclusterbudswereculturedindifferentPH.WhenthepHwasraisedfrom4.01to7.00,thepropagationcoefficientwasrisingatfirst,thendescendinglater.ButthenumbersoffoliarstemofaveragefleshystemwasdescendingatfirstthenrisingwiththepHrising.ThepH6.0wasfavorable.WhentheclusterbudswereculturedinconditionofpH6.0,thepropagationcoefficientwas4.32andtheaveragenumberoffoliarstemperfleshystemwas2.12.IVFrom18to32,thepropagationcoefficientwasrisingwithtemperaturerisingandthenumbersoffoliarstemofaveragefleshystemwasrisingatfirst,thendescendinglater.25wasfavorabletosubculctureofChinesewaterchestnutclusterbuds.Itspropagationcoefficientwas4.32,andaveragenumbersoffoliarstemperfleshystemwas2.12.Thebudsandstemgrowthbestatthistemperature.SothefavorabletemperatureandpHtopropagationofclusterbudswereseparately25and6.0.ThefavorablemediumwasMS+6-BA0.5mg/L+KT0.5mg/L+NAA0.01mg/L+3%sucrose.2Thekindsofplantgrowthregulatorsandtheircompounding,thekindsofdifferentexplants,lightconditionshaddifferenteffectsonthecallusinduction.Theresultindicatedthat24-Dwasthekeyfactorforcallusinduction.Thecytokinin(6-BAKTZT)couldimproveinductedeffectof2,4-D.Itssequencewasthat:6-BAKTZT.ThefavorablemediumtoinducecallusbyusingfleshystemsasexplantswasMS+2,4-D1mg/L+6-BA0.5mg/L+3%sucrose+0.7%agarwith88.19%inductiverate.Theinductedrateoffleshystemswasthehighestoneinallkindsofexplants.Theinductedrateofcallusinthelightpowerof0Luxwashigherthanthatin120Luxor2000Lux.Theresultshowedthatthelowerconcentrationlevelof2,4-Disadvantageoustothepropagationanddifferentiationofcallus.Lingfasuisadvantageoustocallusdifferentiation.TDZhastheverygoodfunctiontomaintainthevigorofcallusandpreventethecallusaging.3TherootinductionofChinesewaterchestnutwaseasy.Theinductedratesoffourmediumswereallhighandtheirdifferencewerenotmarked.Butthedifferenceoftheirrootnumberofaverageplantletwasmarked.Thefavorablemediumwasthat:MS+IBA0.3mg/L+3%sucrose.Itsinductedrateofrootswas100%.Therootsnumberofaverageplantletwas15.(4)Theeffectsofthemethodsofhardening-off,thekindsoftransplantingmedium,thetransplantingenvironmenttothesurvivalrateandthegrowthfettleoftheplantletswerestudied.Theresultsshowedthatthefavorablecombinationofthisthreefactorswasmudofcropland+theculturebottleneedn’tbeopenforsomedaysandtheplantletsweredirectlytransplantedfrombottles+theenvironmentofgreenhouse.(5)ThereisnoreportaboutthecriterionwhichisusedtosystematicallyestimatethecormyieldandqualityofChinesewaterchestnut.Inordertoestimatethecormqualityofthetissuescultureseedlingsandthetraditionalseedlingsforselectingorexploringthegoodstrainbreedinginfurther,theestimatedindexes
